Description The purpose of this module is to introduce participants to the technology currently available to analyse global patterns of gene expression at the protein level. The module will addresss applications of this technology to reveal gene regulation events involved in disease progression as well as potential targets for drug discovery and diagnostics. The module will cover the following aspects of proteomic technologies and research; • Sample preparation from various biological materials (e.g. cell lines, tissue, serum, plasma, etc.) • 2D Gel Electrophoresis, including 2D Difference Gel Electrophoresis (2D DIGE) • Mass Spectrometry (MS) for protein identification and characteristion • Quantitative LC-MS proteomic technologies o SILAC o iTRAQ o Label-free • Introduction to bioinformatics tools for proteomics
Learning Outcomes 1. Select the most appropriate method of sample preparation for various biological materials. 2. Discuss the advantages, limitations and challenges associated with analysis of the proteome by different techniques. 3. Demonstrate knowledge of the differences between the various mass spectrometry techniques for protein identification and characterization. 4. Have an ability to analyze and characterize a protein of interest using various protein bioinformatic software tools.
