Module Aims
|
Module Aims:
- To review the use of enzymes in key industrial processes, both traditional and recently-evolved, with emphasis on their underlying biotechnological strengths and current limitations.
- To outline how biotechnological innovations (especially protein engineering) have resulted in improved products and processes.
- To provide an overview of biosensor development.
- To understand the relevance of DNA recombinant technology for the production & purification of proteins and peptides.
- To learn some important techniques used for the purification and analysis of peptides & proteins.
- To review problems associated with large scale/scale-up production of proteins.

Students will gain:
- An overview of the uses and advantages of enzymes in starch hydrolysis, antibiotic modification and other industrial processes
- An overview of techniques and resources underpinning protein engineering
- An understanding of the potential of protein engineering for the tailoring of useful proteins
- An appreciation of the scope and potential of enzyme and biosensor technologies in the evolution of current, developing and innovative products/processes
- An understanding of the basis of a variety of modern protein separation/analytical methodologies
- An appreciation of the applications of DNA recombinant technology to the production of proteins.

Indicative Syllabus

INDUSTRIAL ENZYMOLOGY
Sources and production of industrial enzymes; enzymes in detergents; starch processing and high-fructose syrup; use of enzymes for antibiotic semisynthesis; leather processing; cellulose degradation in the textile and paper industries.
PROTEIN ENGINEERING
Underlying technologies, incl. databases and bioinformatics; Case history - subtilisin; Insulin and its semisynthesis; Directed evolution of an enzyme.
BIOSENSORS
Definition & applications; biocomponents; transducers; design problems; developments in biosensors incl. examples.
PROTEIN PURIFICATION & ANALYSIS
Advanced Separations (Affinity, Metal-chelate (IMAC), Covalent Chromatographies Purification of Recombinant proteins (His-tag & Thioredoxin fusions , inclusion body formation etc).Genetic Manipulation of protein source for optimal protein/peptide production and stability .
Capilliary Electrophoresis - Protein & Peptide Analysis.
Large scale protein purification and associated problems of scale-up.
Introduction to the concepts of Proteomics and Protein Microarrays
